Hindley grew up in Perth and reached Europe through the amateur ranks, finishing second overall at the Rás in Ireland in 2016 and third at the under-23 Giro d'Italia the year after. He made his Grand Tour debut at the 2018 Vuelta a España, and in 2019 he finished second at the Tour de Pologne, two seconds behind Pavel Sivakov.

The 2020 Giro d'Italia was pushed to October by the pandemic. Hindley won stage 18, from Pinzolo to Laghi di Cancano over the Stelvio, and took the pink jersey two days later at Sestriere, where Tao Geoghegan Hart beat him to the line. The two went into the closing time trial in Milan on identical times, and Geoghegan Hart won the race by 39 seconds.

He abandoned the 2021 Giro in the second week and left for Bora-Hansgrohe. Back in Italy in 2022, he won stage 9 on the Blockhaus, out-sprinting Romain Bardet and Richard Carapaz at the summit, then held station near the top of the classification until stage 20, when he rode away from Carapaz on the Marmolada. He kept the jersey through the Verona time trial, the first Australian to win the Giro.

At the 2023 Tour de France he came off the Col de Marie-Blanque alone to win stage 5 at Laruns and take the yellow jersey, losing it the next day to Jonas Vingegaard and finishing seventh overall. Primož Roglič joined the team for 2024 and Hindley placed 18th at that year's Tour. He rode the 2025 Giro in support of Roglič until a crash on stage 6 near Naples ended his race.

He returned at the 2025 Vuelta a España and finished fourth, thirty seconds behind Tom Pidcock in third. Hindley climbs best on long, steady gradients and can sprint at the top of one, which is how both his Giro stage wins came, at Laghi di Cancano and on the Blockhaus. Against the clock he concedes time to the specialists.
